The thesis covers studying the effect of some cryopreservation factors on structural integrity and functional properties of human blood platelets at low temperature preservation stages. The ability of cryoprotectants of different classes of chemical compounds to scavenge the hydroxyl radicals in model system of their generation and to affect the intensity of LPO processes in the “platelets-plasma” system was investigated. It has been shown that initial morphofunctional state of platelets, compositions of cryoprotective medium and cooling rate of cell suspension within temperature crystallization interval and zone of eutectic temperatures have a crucial effect on the parameters of structural integrity and functional properties of cryopreserved platelets. For the first time it was experimentally proved that use of combination of DMAC with 1,2-PD increased the cryopreservation efficiency of platelets if compared to the application of each cryoprotectant separately. The possibility of effective cryopreservation of platelet concentrates prepared by the buffy coat method and stored for 18 –24 hrs at 22 ± 2 єC was found.
