An effective methods of synthesis of an 3-amino-4-oxo-3,4-dihydroquinazolines derivatives, based on anthranilohydrazide and dicarboxylic acids (oxalic, succinic and phthalic) derivatives were developed.
It was found that proceeding of amide formation is under the influence of NH-acidic properties of diester. It was shown that reactionary character of ethyl ester of 3-succinimdo-4-oxo-3,4-dihydroquinazoline-2-carboxylic acid are defined by spatial factors. It was found that remainder of phthalic acid existed in molecule is defined the way of diacylanthranilohydrazides cyclodehydration. It was shown that interaction of anthranilohydrazide and diethyloxalate in microwave media is accompanied by decarboxylation, different possible ways of this process were considerated.
The simple methods of synthesis of the metyl and ethyl esters of 3-amino-4-oxo-3,4-dihydroquinazoline-2-carboxylic acid. It was first discovered that reactions of oxalamic acid esters with N-nucleophils are proceeding by amide-group and not involving ester-group.
Virtual screening (program PASS) of the synthesized compounds was carried out. Mono- and bis-derivatives of N-(4-oxo-3,4-dihydroquinazoline-3-yl)succinaminic acid are possessing aimed antimicrobial action. The anti-inflammatory and analgesic actions on the rate of test drugs were found for 2-(N’-[2-(3-carboxypropyonylamino)bezoyl]-hydrazinocarbonyl)benzoic acid.
